I have Microsoft Security Essentials installed along with the Comodo firewall with defence + enabled. Would I be well advised to disable either the defence + or the real-time protection of the Microsoft package in case they interfere with each other and if so, does anyone have any useful observations about their relative merits to help me decide which one to kill? Thanks 
MSE is a AV, D+ is a HIPS, they will not interfere becasue they do different things. Just make sure you don’t have two AV’s installed on the computer. IF all you installed was comodo firewall with D+, then all is good.
