If you are a advance user and want to have maximum protection.. Then Yes you should do some tweaking, the normal settings are a bit lowered to not make new users frustrated with too many popups..
Proactive is the highest (built-in) security level. If you want to activate proactive security, open CIS and click Miscellaneous > Manage my configurations.. Pick the "COMODO - Proactive Security" and make sure to press activate, this will result in more pop-ups, but D+ will now monitor and warn you for key-loggers and such.
